C'est l'un des plus grands malentendus.<\/div>\n<div><strong>\"Non dangereux\" ne signifie pas \"sans risque\".<\/strong> - en particulier dans les applications biologiques d\u00e9licates.<\/div>\n<div>Un jour, un nouveau chercheur d\u00e9butant a pris un raccourci : il a pes\u00e9 des LAP \u00e0 l'ext\u00e9rieur d'une armoire de bios\u00e9curit\u00e9 \u00e0 l'aide d'une spatule non calibr\u00e9e. Une l\u00e9g\u00e8re rafale a souffl\u00e9 une minuscule quantit\u00e9 sur sa main. Une erreur d'inattention peut d\u00e9sactiver une bouteille enti\u00e8re.<\/div>\n<div>Dans notre laboratoire :<\/div>\n<ul>\n<li>Les poudres LAP et 2959 sont toujours conserv\u00e9es au r\u00e9frig\u00e9rateur \u00e0 une temp\u00e9rature de <strong>2-8 \u00b0C<\/strong>Les produits de l'agriculture biologique peuvent \u00eatre vendus dans des bouteilles ambr\u00e9es ou envelopp\u00e9s dans du papier d'aluminium.<\/li>\n<li>Les initiateurs au ruth\u00e9nium, plus stables, sont plac\u00e9s dans un dessiccateur \u00e0 temp\u00e9rature ambiante.<\/li>\n<\/ul>\n<div>\n<p>La r\u00e8gle d'or : <strong>Conserver \u00e0 l'abri de la lumi\u00e8re et de la chaleur.<\/strong><\/p>\n<div>Ne les laissez jamais sur un rebord de fen\u00eatre ou au-dessus d'un four.<\/div>\n<\/div>\n<h4>Encore plus important : la gestion des solutions de stock<\/h4>\n<div>\n<p>Une erreur fr\u00e9quente chez les nouveaux \u00e9tudiants :<\/p>\n<div>Ils pr\u00e9parent un grand lot de solution LAP, en d\u00e9cong\u00e8lent une partie pour l'utiliser, puis recong\u00e8lent le reste. Apr\u00e8s plusieurs cycles de cong\u00e9lation-d\u00e9cong\u00e9lation, l'hydrogel ne se r\u00e9ticule plus correctement.<\/div>\n<\/div>\n<div>Pourquoi ? Les cycles de gel-d\u00e9gel d\u00e9truisent l'activit\u00e9 des photo-initiateurs.<\/div>\n<div>\n<p>Ma recommandation :<\/p>\n<div><\/div>\n<p><strong>Aliquotez vos solutions de base.<\/strong><\/p>\n<div>Utiliser des tubes Eppendorf st\u00e9riles, divis\u00e9s en volumes \u00e0 usage unique (par exemple, 50 \u03bcL ou 100 \u03bcL), utiliser une fois, jeter. Pour la culture cellulaire, de petites diff\u00e9rences de concentration peuvent affecter consid\u00e9rablement la viabilit\u00e9.<\/li>\n<li><strong>Dissolution<\/strong>:\n<ul>\n<li><a href=\"https:\/\/changhongchemical.com\/product\/photoinitiator-lap-cas-85073-19-4\/\"><strong>LAP<\/strong><\/a>: Dissoudre directement dans du PBS 1\u00d7 ou du milieu cellulaire. Pour les cultures \u00e0 long terme, j'ajoute souvent 1% <strong>p\u00e9nicilline-streptomycine<\/strong> pour une protection suppl\u00e9mentaire.<\/div>\n<div><\/div>\n<hr \/>\n<h3>Meilleures pratiques dans le monde r\u00e9el : Protocoles pour diff\u00e9rents sc\u00e9narios<\/h3>\n<h4>Sc\u00e9nario 1 : bio-impression 3D<\/h4>\n<div>L'application la plus populaire - et la plus difficile -.<\/div>\n<div><\/div>\n<ul>\n<li>\n<div>\n<p><strong>La concentration est un exercice d'\u00e9quilibre<\/strong><\/p>\n<div>Pour le LAP, une fen\u00eatre de travail typique est <strong>0,03% - 0,5% (w\/v)<\/strong>.<\/div>\n<div>Trop faible : r\u00e9ticulation incompl\u00e8te, effondrement de la structure.<\/div>\n<div>Trop \u00e9lev\u00e9 : cytotoxicit\u00e9.<\/div>\n<div>Optimisez pour votre hydrogel et votre type de cellule. En cas de rougeur, consultez un m\u00e9decin et apportez les informations sur le r\u00e9actif.<\/li>\n<li><strong>Exposition des yeux<\/strong>: Urgence ! Utilisez une douche oculaire ou du s\u00e9rum physiologique, <strong>maintenir les paupi\u00e8res ouvertes, rincer continuellement pendant au moins 15 minutes<\/strong>Si vous avez des probl\u00e8mes de sant\u00e9, rendez-vous imm\u00e9diatement \u00e0 l'h\u00f4pital.<\/li>\n<li><strong>Inhalation<\/strong>: Transporter la personne \u00e0 l'air frais.
